In a game that has a singleplayer and multiplayer component, like Call of Duty, would it be right to force the players to play the singleplayer to unlock multiplayer. Like, have multiplayer locked until players compoets the story mode or reach a certain point in it?

Depends... I think in StarCraft 2 this should be, because you need to have some practice in single player mode before attempting multi player... But in something like MineCraft... nope.

Depends on the game. If it's a game that requires a tutorial, not your standard FPS, then maybe. However, I bought Battlefield: Bad Company 2 only for the multiplayer. In fact, I don't think I ever touched the single player yet. I still don't think a single player playthrough should be required though, maybe an intense tutorial? In the end, it's usually best to give the player as many options as possible.

i must say no, because locking such a thing would simply make me frustrated, i want to play games in my pace, not others, same goes for the SC2 thingy, if i want to go straight into the MP, then i go straight into the MP, especially if it is one of the stronger parts of the game..
