I got this off of the PCStats newsletter (I highly recommend it, if only for weekly tech tips) and it cut the loading times I experience for my game (havn't tried others so far) into 1/4th of what it used to be.

Here it is:

Not all computers are created equal, and so Microsoft has had to try and write their OS's for the masses, meaning Windows may not be optimized very well for your PC. Luckily, Microsoft also built in a few switches that help you to fine tune your system.

Today we're going to optimize Windows File system Caching which should hopefully improve performance of your system a bit. First load up regedit (Start, Run then type regedit and press ok) and follow this path. HKEY_LOCAL_MACHINE -> SYSTEM -> Current ControlSet -> Control -> Session Manager -> Memory Management.

From here find the IoPageLockLimit and change its value according to how much system memory you have.
4096 - 32MB or less of memory
8192 - 32MB+
16384 - 64MB+
32768 - 128MB+
65536 - 256MB+

After that's done save and reboot. Now that Windows has been tuned for your OS you should notice a bit of a speed increase, especially when dealing with memory intensive programs. =)

That's for Win 2k/NT/XP only. If your run 98 you would have to change the system.ini. Add or change the section

[vcache]
MaxFileCache=xxxx

replace xxxx with a value from Ash's table.

Then add it
In XP in the registry.

Yeah right, I don't mess with the registry unless I know what I'm doing, thus I never mess with it.

Someone could make a little .REG add in for the registry for you. Then you'd simply have to double click it and add it. The registry is only scary because M$ are soooo damn disorganised, it's really just a database of settings.

Yeah, I'm also running XP Pro and don't have a IoPageLockLimit key. Would be a great tip though.
