One of two ways.

1. Check the coordinates of the mouse against the sprite

or

2. Position a 1x1 hidden sprite at the mouse coordinates and check for collision

[EDIT]You can only change the DBC icon using an external program on the final exe - that's all I know on that subject, other people can guide your further.

to change the icon, simply make a shortcut to your game, right click, properties, and change the icon image

That changes what the icon looks like just on your desktop only - not the one associated to the exe file, so when you give your exe to someone else, it will still be the standard DB grey square icon.

To change it in the exe itself so everyone who downloads it sees it, then you have to use IanM's method - using a separate program like Icon Commander.
